Applications of nucleoside-based molecular probes for the in vivo assessment of tumour biochemistry using positron emission tomography (PET) BABT
Wiebe,Leonard I..
Positron emission tomography (PET) is a non-invasive nuclear imaging technique. In PET, radiolabelled molecules decay by positron emission. The gamma rays resulting from positron annihilation are detected in coincidence and mapped to produce three dimensional images of radiotracer distribution in the body. Molecular imaging with PET refers to the use of positron-emitting biomolecules that are highly specific substrates for target enzymes, transport proteins or receptor proteins. Molecular imaging with PET produces spatial and temporal maps of the target-related processes. Molecular imaging is an important analytical tool in diagnostic medical imaging, therapy monitoring and the development of new drugs. Molecular imaging has its roots in molecular biology....

Approche de l'état physiologique de l'huître creuse Crassostrea gigas au cours d'un cycle saisonnier à Marennes-Oléron ArchiMer
Moal, Jeanne; Samain, Jean-francois; Bodoy, Alain; Le Coz, Jean-rene.
The energy charge was measured for two Crassostrea gigas populations, during an annual survey at Marennes Oleron, to appreciate the condition of health of these oysters. It is assumed that the actual decrease in production of this area is related to a biomass overload which affects the oysters by lowering their scope for growth. Energy charge, an index developped by ATKINSON (1968), defines the energy equilibrium of organisms. It is the adenylic nucleotide ratio: E.C = (ATP + 1/2ADP)/ATP + ADP + AMP). We have measured this parameter to determine the state of health of these oysters. A methodological study has been conducted previously to define the conditions of sampling, conservation and extraction for a HPLC assay. The HPLC method (high performance...

Pelícia,VC; Sartori,JR; Zavarize,KC; Pezzato,AC; Stradiotti,AC; Araujo,PC; Mituo,MAO; Madeira,LA.
This study aimed at evaluating the effect of nucleotides on the performance and carcass yield of broilers fed diets with no antibiotic growth promoters (AGP), anticoccidials, or animal feedstuffs. In the trial, 600 Ross 308 male broilers were distributed in a completely randomized experimental design into six treatments with four replicates of 25 birds each. Treatments consisted of a control diet (CD), CD + AGP, CD + 0.04%, CD + 0.05%, CD + 0.06%, and CD + 0.07% nucleotides. The experimental diets did not contain anticoccidials, and birds were vaccinated against coccidiosis at three days of age. No significant differences were detected among broilers submitted to the different treatments in none of the studied parameters. Under the conditions of this...

Improvement in RNA extraction from S. cerevisie by optimization in the autolysis and NH3 hydrolysis BABT
Oliveira,Antonio Martins; Oliva Neto,Pedro de.
The optimization of autolysis of Saccharomyces cerevisiae from brewery was studied aiming at the maximum ribonucleic acid extraction and yeast extract production. The best conditions for yeast autolysis was 55.2ºC, pH= 5.1 and 9.8% NaCl for 24h of processing, without the NH3 use. In these conditions, the RNA yield was 89.7%, resulting in 51.3% of dehydrated yeast extract with 57.9% protein. The use of 12.2% NH3 at 60ºC after autolysis (8h) and plasmolysis (8h) was not viable due to the reduction in the RNA yield from 89.7to78.4%. On the other hand, the thermal shock at 60ºC for 15 minutes prior to autolysis provided an increase in the yield from 89.7 to 91.4%. The autolysis, including NaCl plasmolysis in the optimized conditions was efficient, economic and...

La detection des effets subletaux des pollutions: Utilisation in situ d'un indice biochimique, la charge energetique ArchiMer
Sylvestre, C; Batrel, Y; Legal, Y.
Marine water management implies the provision of environmental evolution. This can be seen with tests giving information on organism health. After a presentation of biochemical parameters (energetic charge) the authors describe the technique used and the results obtained with Nassarius reticulatus and Crangon crangon . In practice, the evolution of ratio of energetic charge to total nucleotides is a signal of imbalance in the ecosystem.

Oyster adenytate energy charge: response to levels of food ArchiMer
Moal, Jeanne; Le Coz, Jean-rene; Samain, Jean-francois; Daniel, Jean-yves; Bodoy, Alain.
Adenylatc Encrgy Charge (AEC) values of Crassostrea gigas (9 and 18 months old) cultured in ponds in Marennes-Oléron Bay (Atlantic coast, France) are reported. Two ponds were loaded with two different initial oyster biomasses in order to obtain different food conditions. The growth of oysters was different in the two ponds which corroborated the effectivences of attaining different food conditions. AEC level was different depending on the pond and was therefore related to food availability per unit oyster biomass. Well-fed oysters exhibited greater AEC values than poorly-fed oysters, throughout thc study. Information on nutritional situations for wild or cultured populations can therefore be obtained by AEC. In cach pond, the seasonal AEC variations were...

Ribonuclease Production by Aspergillus species Rev. Microbiol.
Gomes,Eleni; Silva,Roberto da; Serzedello,Alcides.
Ribonuclease production by Aspergillus flavipes, A. sulphureus and A. fischeri in semi-synthetic medium, after 24-144 hours at 30ºC under shaking, was studied. After cultivation, the medium was separated from micelia by filtration and the resultant solution was used as enzymatic extract. The highest amount of biomass and RNase was obtained after 96 hours of cultivation. The enzymes produced by three species presented similar characteristics, with optimum temperature at 55ºC and two peaks of activity at pH 4.5 and 7.0. A. flavipes RNases were more sensitive to temperature: 50% of the initial activity was lost after 1 hour at 70ºC. After this heat treatment, RNase of A. sulphureus lost 30% of this activity and that of A. fischeri only 16%. The nucleotides...

Sources of trophic action on performance and intestinal morphometry of broiler chickens vaccinated against coccidiosis Rev. Bras. Ciênc. Avic.
Sakamoto,MI; Faria,DE; Nakagi,VS; Murakami,AE.
The aim of this experiment was to evaluate the effect of sources of trophic action (glutamine alone, glutamine associated with glutamic acid and yeast) associated with vaccination against coccidiosis on the performance and morphometry of the small intestine of broilers. In the trial, 1,200 broiler chicks were distributed according to a completely randomized design with a 3 x 2 + 2 (trophic action x vaccination or not + control - free trophic factor) factorial arrangement, with five replicates of 30 birds each. Vaccination negatively affected performance parameters and the morphometry of the intestinal mucosa, but at the end of the experimental period, among the broilers vaccinated against coccidiosis, the group fed glutamine presented better recovery from...

Use of nucleotides in weanling rats with diarrhea induced by a lactose overload: effect on the evolution of diarrhea and weight and on the histopathology of intestine, liver and spleen BJMBR
Norton,R.; Leite,J.; Vieira,E.; Bambirra,E.; Moura,C.; Penna,G.; Penna,F..
Until recently, dietary sources of nucleotides were thought not to be essential for good nutrition. Certain states with higher metabolic demands may require larger amounts that cannot be provided by endogenous production. The objective of the present study was to determine the action of nucleotides on the recovery from lactose-induced diarrhea in weaned rats. Thirty-six weanling Fisher rats were divided into two groups. Group 1 received a standard diet and group 2 received a diet containing lactose in place of starch. On the 10th day, six animals per group were sacrificed for histopathological evaluation. The remaining animals were divided into two other subgroups, each with 6 animals, receiving a control diet, a control diet with nucleotides (0.05%...

Yeast extract with blood plasma in diets for piglets from 21 to 35 days of age R. Bras. Zootec.
Pereira,Cinthia Maria Carlos; Donzele,Juarez Lopes; Silva,Francisco Carlos de Oliveira; Oliveira,Rita Flávia Miranda de; Kiefer,Charles; Ferreira,Aloízio Soares; Hannas,Melissa Izabel; Brustolini,Paulo César.
The objective of this study was to evaluate the inclusion of yeast extract as a partial replacemer of blood plasma in piglet diets and its effect on the performance and intestinal morphometry of pigs weaned at 21 days of age. One hundred and twenty animals were randomized into blocks, with five diets (4.0% blood plasma; 2.0% blood plasma with 0.0; 1.0; 2.0 or 3.0% yeast extract), six replicates and four pigs per experimental unit. At 35 days of age, one pig per experimental unit was slaughtered in order to evaluate duodenal and jejunal morphometry. The levels of yeast extract had a quadratic impact on daily feed intake and final weight, which increased up to the estimated level of 1.91%. Daily weight gain and feed conversion rate were not affected. A...
